Platelet Rich Plasma or PRP is derived from the patient’s blood. Processing the patient’s blood specimen in a centrifuge separates the blood components and concentrates the platelets in the plasma. The concentrated platelets found in PRP are rich in growth factors along with bioactive proteins that are vital to initiate and accelerate tissue repair and regeneration. These bioactive proteins increase stem cell production to initiate connective tissue healing, bone regeneration and repair, promote development of new blood vessels, and stimulate the wound healing process.

In PRP, platelets are not active that is platelets are not ready to release growth factors from their organelle named Alpha-Granule, while in PRF, platelets become activated due to the addition of calcium chloride and thus provide immediate results at the site of injection.

In PRP, after injection, platelet could be moved from the site of injection to the other area, thus visible effect will not be attained at the site of injection, while in PRF, fibrin matrix holds platelets firmly at the site of injection thus growth factors are released at the desired area for longer period of time.

Prizmah is better than other PRP kits because;

  • Prizmah removes almost all of the RBC in the PRP, thus Prizmah reduces the risk of hemosiderin staining & thus cell injury.
  • Prizmah removes almost all of the WBC in PRP. Thus by removal of WBCs from plasma, Prizmah reduces the risk of oxygen radicals’ production, inflammation, impaired healing & local pain.
  • Prizmah has built in calcium chloride in exactly the right concentration to initiate fibrin polymerization to create a matrix to keep the platelets at the site of injection and to create a scaffold to support cell migration (stem cells, fibroblasts) and for sustained growth factor release.
  • Prizmah is easier to use, takes less time, requires less blood and is not user dependent like other systems. The blood and PRP is also not exposed to air/oxygen during the process, I.e. Closed system.
  • The anticoagulant in Prizmah is sodium citrate which has a close to physiologic pH (7.4).

Prizmah is a patented technology for the safe and rapid preparation of autologous Platelet-rich Fibrin, using a small amount of a patient’s own blood.

Procedure

A safe, simple-step process:

  • 9 cc peripheral blood is drawn into a blood collecting tube.
  • This tube is then spun in a centrifuge for 6 minutes at 1100 g or 2800 rpm, separating the Platelet-rich Plasma (PRP) from other elements of the blood. After centrifuge, this tube has clearly three layers, upper layer of pure PRP, middle layer of separator gel with WBCs and lower layer of RBCs.
  • The PRP is then transferred into an activation tube already has an activator (Calcium Chloride) through a safe, closed system. Here PRP is converted into Platelet-rich Fibrin (PRF) due to Calcium Chloride.
  • 3 to 4 times up-down mild rotation of activation tube is also required at this point of time so that Calcium Chloride mixes thoroughly in PRP and to convert it into PRF uniformly.
  • PRF is then injected into the treatment areas recommended by physician.
  • It must be noted that PRF should be injected within 20 to 25 minutes after its preparation, because as the time passes PRF becomes more & more viscous due to the presence of fibrin matrix and physicians may experience very hard to inject it at desired area (specially in facial skin).
  • Always Unlock the lid by pressing the ‘OPEN / STOP’ button.
  • Close and latch the lid. The lid is properly latched when the yellow ‘LATCH’ indicator light is illuminated.
  • Turn the centrifuge on by pressing the red ‘START’ button
  • For proper centrifugation and getting PRP, Leveling of Centrifuge from the base is very much necessary.
  • Always put Centrifuge on leveled, flat & hard surface to avoid interruption in centrifugation and improper PRP separation.

Centrifuge must contain a balanced load to work properly. Use the following rules when loading the rotor;

  • Opposing tube holders must be identical & must contain the same cushion.
  • Opposing tube holders must be empty or loaded with equally weighted samples.

Please note that after 6 min, if there is an improper PRP separation than another 6 minutes centrifugation is required.

Platelet Rich Plasma or PRP has got after processing the patient’s blood specimen in a centrifuge for certain period of time which separates the blood components and concentrates the platelets in the plasma. While if any activator or fibrin forming agent like thrombin or Calcium Chloride is added into PRP, fibrin threads start to appear in the form of matrix or net, now PRP with activated Platelets is known as Platelet Rich Fibrin Matrix.

The body signals platelets and other components in our blood supply to migrate to the site of injury. Platelets are the injury’s first responders, and under normal conditions these platelets release various factors that initiate and subsequently stimulate skin & tissues regeneration and repair, promote new blood vessel development to accelerate tissue regeneration, and stop the bleeding. Platelets release many other bioactive proteins responsible for attracting macrophages, mesenchymal stem cells, and osteoblasts.
Inside the platelet are 2 types of granules, namely, alpha granules and dense bodies. Alpha granules contain the clotting and growth factors that are released in the healing & regeneration process. Normally at the resting state, platelets require a trigger to activate and become a participant in regeneration, wound healing and hemostasis. This activation is provided by addition of Calcium Chloride in PRP to convert it in PRF.

  • Safety blood collection set with butterfly wing – vacuum system for blood drawing prevents contamination of air or oxygen during process.
  • Blood collecting tube which is specially modified to collect just 9 cc whole peripheral blood. This tube also contains patent Thixotropic Separator Gel which perfectly separates RBCs & WBCs from the PRP after 6 minutes’ centrifugation at 1100 g or 2800 rpm. Furthermore, it also contains Sodium Citrate as anti-coagulant. The benefit of Sodium Citrate is to maintain the natural pH of blood in PRP and PRF that 7.4, thus minimizes the pain or injury at the site of injection.
  • Jacketed Device (closed system) with protection sleeve for the sterile transfer of PRP into PRP collection tube or activation tube.
  • Activation tube which contains Calcium Chloride as Fibrin Forming solution (activator). Other PRP kits use bovine thrombin which may cause serious allergic reaction at the site of injection.
  • PRP collection tube. In this tube PRP can be kept for almost 4 hours after maintain the temperature (3-4 0C).
